A killer is on the run as police probe the fatal shooting of a man overnight.

Officers were called to the inner-Melbourne suburb of Ascot Vale about 12.15am on Sunday after a man in his 30s was found with suspected gunshot wounds.

He was rushed to hospital in a critical condition but did not survive.

Homicide Squad detectives were among the numerous police still at the scene by late Sunday morning.

Officers were blocking North Street to traffic as they searched the area for clues.

Forensic officers were seen removing blood-covered clothing from the scene, while bloodstains were also visible on the ground.

Witnesses say they heard numerous gunshots ring out in the area just after midnight.

It is believed the shooting took place on Mount Alexander Road before the injured man made his way to a supermarket loading dock in nearby North Street, where he was found by emergency crews.

Police have called for any witnesses to come forward.
